چکیده

Studies have shown the prognostic significance of disseminated tumor cells (DTC) in bone marrow of patients with colorectal cancer. However, the molecular characteristics of DTCs, including their miRNA expression profiles, remain mostly unknown. In this study, we analyzed the miRNA expression of DTCs in bone marrow. EpCAMþ bone marrow cells were collected using immunomagnetic beads after exclusion of CD14þ and CD45þ cells, then subjected to miRNA microarray analysis. Cluster analysis (7 colorectal cancer patientswith livermetastasis and 12 colorectal cancer patientswithout livermetastasis) indicated thatmiR-340 andmiR-542-3p expressions were significantly decreased in EpCAMþ bonemarrow cells of patients with liver metastasis (P 1⁄4 0.019 and 0.037, respectively). We demonstrated that pre-miR-340 administration inhibited growth of colon cancer cells and suppressed c-Met expression in vitro. In clinical samples of colorectal cancer, miR-340 was expressed at significantly lower levels in tumor tissues compared with normal mucosa. Survival analysis in 136 patients with colorectal cancer indicated that low miR-340 expression was correlated with shorter 5-year disease-free survival (P1⁄4 0.023) and poor 5-year overall survival (P1⁄4 0.046). It was of note that the colorectal cancer group with low miR-340 and high c-Met expression had the worst prognosis. We further demonstrated that systemic pre-miR-340 administration suppressed growth of pre-established HCT116 tumors in animal therapeuticmodels. These findings indicate thatmiR-340may be useful as a novel prognostic factor and as a therapeutic tool against colorectal cancer. Our data suggest that miR-340 in bone marrow may play an important role in regulating the metastasis cascade of colorectal cancer.Mol Cancer Ther; 13(4); 976–85.

Decreased miR-155-5p, miR-15a, and miR-186 Expression in Gastric Cancer Is Associated with Advanced Tumor Grade and Metastasis

Background: Gastric cancer (GC) is one of the most prevalent cancers with a high rate of mortality in the world. In recent years, microRNAs (miRNAs) have been proposed to be involved in GC development. In this study, we aimed at investigating differential expression level of miR-155-5p, miR-15a, miR-15b, and miR-186 in GC. Methods: For this research, we used qPCR to investigate miR-15b, miR-155...

نقش microRNA-21 در سرطان کولورکتال و قابلیت آن به عنوان یک بیومارکر پیش بینی کننده، تشخیصی و پیش آگهی: مقاله مروری

Colorectal cancer (CRC) is one of the most common types of cancers worldwide. Despite the improved therapeutic and diagnostic strategies and screening programs, morbidity and mortality of CRC is still considerable. Currently, most common approaches for diagnosis of CRC are colonoscopy and fecal occult blood test (FOBT). Because of the invasive nature of colonoscopy and low sensitivity of FOBT, ...

Investigation of MicroRNA-21 Expression Levels in Serum and Stool as a Potential Non-Invasive Biomarker for Diagnosis of Colorectal Cancer

Background: Most cancer studies focus on exploring non-invasive biomarkers for cancer detection. In the present study, we sought to investigate the expression level of microRNA-21 (miR-21), as a potential diagnostic marker, in serum and stool samples from 40 patients with colorectal cancer (CRC) and 40 healthy controls. Methods: Quantitative real-time RT-PCR was applied to determine the relativ...

بررسی نقش miR-429 با میزان بیان ژن E2F3در رده سرطان پستان

Background and purpose: MicroRNA regulation of cancer-related pathways has a major effect on cancer development. MiR-429 is a member of miR-200 family with an important role in metastasis. There are many studies on the association between miR-429 and liver, bladder, bowel, and gastric cancers. In this study, we investigated the expression changes of E2F3 genes after transfection of MCF-7 cells ...
